Hybridization of polynucleotides on functionalized self-assembled monolayer

چکیده

For the hybridization of single-stranded polynucleotides at a solid-liquid interface without any modifications of the polynucleotides, a self-assembled monolayer (SAM) containing anthryl groups which have an affinity for nucleic acid bases was prepared. The hybridization was investigated by surface plasmon resonance (SPR) and atomic force microscopy (AFM). Results of SPR measurement indicated that hybridization of polynucleotides occurred on the SAM. The AFM images of the SAM showed that a network structure was induced by the polyadenylic acid (poly(A)) and polyuridylic acid (poly(U)) or polyguanylic acid (poly(G)) and polycytidylic acid (poly(C)) hybridization. These results suggested that hybridization of polynucleotide occurs at the monolayer level.
